Collagen-based double layered biomimetic tubular materials for vascular and tracheal applications

Elaborating tubular grafts is challenging. Despite the high clinical demand for vascular implants of limited diameter and for tracheal grafts, few solutions in the market cover the necessary features for successful implantation. Synthetic, low diameter vascular grafts are known for their limited endothelialization and reduced patency. In the case of the trachea, the most promising solution is to replace it with cryopreserved aortas rather than using synthetic materials, as demonstrated by Martinod et al. (1,2). An alternative strategy relies on biomimetic scaffolds, as they show low immune response, excellent biocompatibility and potential regeneration. Type I collagen is the prevalent component of the extracellular matrix it is often selected as the protein of choice to engineer tissue analogs. Our approach combines the previously reported tubular materials produced by ice-templating and topotactic fibrillogenesis (3,4,5) to which we now add a dense inner collagen layer. These crosslinker-free materials reproduce the multi-scale architecture of native tissues and reach mechanical properties (radial compliance up to 6%, longitudinal elastic modulus up to 30 kPa in quasi-static conditions) and water-tightness (resisting up to 200 mmHg luminal pressure) relevant for in vivo tracheal implantation. The bare materials were imaged by TEM to assess collagen ultrastructure and validated by surgeons for suturability. Upon seeding with hMSCs and HUVECs extensive cell colonization and endothelium formation were observed. In summary, our approach opens an exciting pathway for obtaining a family of off-the-shelf grafts applicable both in vascular and tracheal conditions.
