Résumé

The emergence of bacteria that are resistant to multiple drugs is a significant clinical obstacle for the treatment of infections worldwide. Pseudomonas aeruginosa is a type of Gram-negative bacteria that can cause infections in hospitals around the world and severe infections in individuals with cystic fibrosis. Colistin is an antimicrobial peptide that disrupts the outer membrane of Gram-negative bacteria, causing cell lysis, and is a key component of this project. Since it is toxic for human and veterinary health, research about this antibiotic lost its attention for a while. But it gained attention again in the 1990s due to its effectiveness against infections caused by multi-drug resistant Gram-negative bacteria. However, resistance to this antibiotic is increasingly becoming more common. As a result, a new approach is emerging to identify other agents that can work in combination with colistin to restore its efficacy. This project aims to investigate the mechanisms behind the synergistic antibacterial activity of a recently approved DrugA in combination with colistin in P. aeruginosa by using advanced fluorescence microscopy techniques along with genetic and cell biology approaches.
