Monitoring of de novo lipogenesis in vivo via palmitic acid labelling with deuterium

Résumé

De novo lipogenesis is a metabolic pathway allowing the de novo synthesis of fatty acids in a large number of organs such as liver, muscle and adipose tissue. This pathway can be altered in several human diseases. Its quantification can help to better understand and characterise the pathological mechanisms involved. There are several non-invasive methods using deuterated water (2H2O) and 2H incorporation into palmitic acid (C16:0) to determine lipogenic flux in vivo. This is what we have tested and developed in mice in this project. After ingestion of 4% deuterated water and sacrifice of mice, total fatty acids were extracted from different tissues (plasma, liver, adipose tissue) via Bligh and Dyer extraction followed by basic hydrolysis. After derivatisation with pentafluorobenzene, the extracts were analysed on a negative chemical ionisation GC-MS. The raw data were reprocessed in the IsoCor software, which removes the natural abundance of 13C and calculates the average enrichment obtained. Then the relative abundance of palmitic acid and its isotopologues was calculated via an area ratio with the internal standard (C19:0 acid). This method was set up and tested on mice with a lipogenesis defect (Chrebp knock-out mice) and then on a mouse model of obesity with increased lipogenesis. This study highlights a simple and robust method for studying lipogenesis by reaching the steady state of deuterium incorporation after 3 weeks of 2H2O administration.
