HARD TICKS IDENTIFICATION USING MALDI-TOF MS: CONSTRUCTION OF A MAIN SPECTRA LIBRARY OF IXODID TICKS FROM URBAN WILDLIFE

Résumé

A total of 108 hard ticks were collected from 85 European hedgehogs, 17 roe deers and 6 red fox. Morphological features allowed the identification of 79 I. hexagonus and 33 Ixodes ricinus 1. Most ticks were females (fig. 2). At least 4 specimens of each species were used for the creation of the Main Spectra Library (MSP), using the FlexControl and FlexAnalysis software (Bruker Daltonics). Species identification was confirmed by qPCR (ITS2 gene) using the tick's bodies selected for the MSP 2. At least 12 additional ixodid ticks were subjected to MALDI-TOF MS analysis. 79% of samples had good-quality spectra. A comparative analysis of MS spectra for intra-species reproducibility and inter-species specificity was performed. The visualization of two MS spectra per tick species showed perfect intra-species reproducibility and inter-species specificity (Fig. 3A). Similarly, the dendrogram made with the MSPs from both species showed that specimens of the same species grouped on the same branch, confirming intra-species reproducibility and interspecies specificity (Fig. 3B). The spectra of the same species showed a high level of similarity, with the CCI value 1.00 (Fig 3C). I. hexagonus displayed CCI within the range of 0.57 to 0.76 while it was between 0.05 and 0.82 for I. ricinus. The CCIs between different species displayed lower values confirming the possibility for spectra species level discrimination.
