Resetting naïve state features with epigenetic inhibitors in rabbit iPS cells

Résumé

Pluripotent stem cells (PSCs) exist in two distinct states of pluripotency called ‘naïve’ and ‘primed’. These two states differ in signaling pathways, growth factor dependence, cell cycle duration, and epigenetic regulation. Moreover, only the naïve PSCs can make systemic chimeras after injection into preimplantation embryos. Unlike rodents, rabbit PSCs only exist in the primed state. The objective is to replace Vitamin C, combining epigenetic inhibitors (BRD4354 (class II histone deacetylase inhibitor), A366 (histone methyltransferase G9a inhibitor), and UF010 (class I histone deacetylase inhibitor)) to ALGöX medium in order to promote chromatin opening and allow “naïve” reprogramming factors action.
