The present invention relates to a method for estimating the distribution of particle response times of a system, in particular for the purpose of characterising said particles. The method is applicable, for example, to time-resolved fluorescence measurements. The method at least comprises: a step (31) of determining a function approximating the log-likelihood of the target distribution ?o corresponding to a distribution with no pile-up effect based on measurements of response times of particles detected with a pile-up effect, that is, measurements of the distribution g?o; a step (32) of maximising the approximated log-likelihood of all the parameters (?) in order to determine the estimator as the vector ? that maximises the approximated log-likelihood.
