Fluorinated Triazole Foldamers: folded or extended conformational preferences

While on the one hand the field of peptide-based foldamers capable of providing mimics of secondary structures has aroused growing interest particularly in the field of protein-protein interactions, and on the other hand fluorinated drugs have taken hold in the pharmaceutical market, fluorinated peptidomimetic foldamers are still in their infancy. We report here the easy access to fluorinated triazolamers based on 2-amino-3,3,3-trifluoropropyl-1,4-triazolyl acetic acid (CF3-1,4-Tz) and on aminomethyl-1,4-triazolyldifluoroacetic acid (1,4-Tz-CF2). Their conformational analyses by spectroscopic and computational analyses revealed that they are capable to adopt short multi-stranded β-sheet like and elongated βstrand like structures, respectively.
