Résumé

The aim of this project was to improve knowledge on the antibacterial activity of the synthetic compound CC20, an analogue of the Cytosporone E. Preliminary results showed that CC20 exhibits good antibacterial activity against MRSA (MIC = 1.25 μ g/mL) but has no effect against Pseudomonas aeruginosa or Escherichia coli strains (MIC > 150 μ g/mL) [1]. The first objective was to find out if efflux pumps and/or membrane impermeability play a role in the resistance mechanism of Gram-negative bacteria. For this, the MIC of the CC20 has been determined against 9 efflux pump mutants of E. coli BW25113 and permits to highlight that TolC channel was essential in the resistance (MIC = 5 μ g/mL) of E. coli. Furthermore, membrane impermeability has not been shown to be implicated in the resistance of E. coli to CC20. Spontaneous mutation frequency was performed on the E. coli Δ tolC strain against CC20 and no resistance mutant against the molecule was obtained (Frequency < 10-13). The second objective was to work on the characteristics of CC20 against MRSA. We highlight that the strain used has a low frequency of resistance to CC20, (< 10-13); and the bactericidal activity is visible around 12 hours for 25 μ g/mL of CC20, corresponding to 20 times the MIC.
