Une modification de l'environnement embryonnaire influence le phénotype des descendants sur trois générations chez la caille

The epigenetic phenomena, such as the DNA methylation, which participate to the regulation of gene expression,
can influence the phenotypes and recent studies show that the epigenetic information can be transmitted across
generations. In the context of the ANR research program EpiBird, we conducted a pilot study to estimate the
phenotypic differences in the third generation (G3) between two lines of quails, issued from the same ancestors
(G-1), obtained by treating the eggs (G0 Epi+) or not treating them (G0 Epi-) by injection of genistein. The
genistein interferes with the epigenome (especially with the DNA methylation) and is a phyto-estrogen known
for its impact on the reproductive system in several species, at least in the first generation of descendants. After 3
generations without treatment, a significant difference in the sexual maturity of animals, which is no longer
attributable to direct maternal effects, was observed between the lines Epi + and Epi-: the Epi-animals started
earlier to lay eggs and produced more eggs throughout their laying career. A significant interaction between line
and sex was observed for the body weight at 3 weeks of age. The results of this pilot study are highly valuable in
the analysis of the multi-generational impact of the environment on the phenotype of livestock.
Les phénomènes épigénétiques, comme la méthylation de l'ADN, en participant à la régulation de l'expression
des gènes, peuvent avoir une influence sur les phénotypes. Des études montrent en outre que l'information
épigénétique peut être transmise à travers les générations. Dans le cadre du programme ANR EpiBird, nous
avons réalisé une étude pilote afin d’estimer les différences phénotypiques dans la troisième génération (G3)
entre deux lignées de cailles issues des mêmes ancêtres (G-1), aux oeufs traités (G0 Epi+) ou non (G0 Epi-) par
une injection de génistéine. La génistéine perturbe l'épigénome (notamment au niveau de la méthylation de
l'ADN) et est un phyto-oestrogène connu pour son impact sur l'appareil reproducteur dans plusieurs espèces, au
moins sur la première génération de descendants. Après 3 générations sans traitement, une différence
significative sur la maturité sexuelle des animaux, qui n’est alors plus attribuable aux effets maternels directs, a
été observée entre les lignées Epi+ et Epi- : les animaux Epi- sont plus précoces et pondent plus d’oeufs sur leurs
carrières. Un effet significatif a également été démontré sur l’interaction entre le poids à 3 semaines et le sexe
des animaux. Les résultats de cette étude pilote sont d'ores et déjà très intéressants dans l'analyse de l'influence
multigénérationelle de l'environnement sur le phénotype des oiseaux d'élevage.
