A deletion in a photosynthesis-related gene (PpPSAK) determines the anthocyanin accumulation in the mesocarp of French blood-flesh peaches

Résumé

Little is known about the mechanisms by which light controls anthocyanin biosynthesis in fruit flesh. Yet the accumulation of anthocyanin pigments is a major determinant of peach (Prunus persica L. Batch) fruit quality. We studied the recessive blood-flesh trait (bf), identified in French cultivars by mapping and then localized on chromosome 4. On genetic, molecular and transcriptional grounds, we identified the underlying gene linked to this bf trait. A metabolomic study showed that blood-flesh cultivars had high levels of cyanidin-3-glucoside and cyanidin-3-rutinoside in the fruit mesocarp around 45 to 60 days after anthesis. Fine mapping coupled with a positional cloning strategy mapped the bf locus to an 80 kb interval on peach linkage group (LG) 4. Sequence polymorphism analysis of the genes present in this interval identified a single candidate gene for bf as a 21-bp deletion in exon 3 of a PSAK-type gene. A specific marker for the deletion was developed and validated in various genetic backgrounds. A comparative transcriptomic analysis of fruit flesh from a bf and non-bf cultivar at 4 stages of fruit development, in kinetics ranging from 60 days after flowering to fruit ripening, revealed co-expressed genes. Overexpression of PpPSAK in the flesh and midrib of bf fruit correlated with the expression of structural anthocyanin biosynthesis genes. Protein-protein interactions were observed and functional pathways were discussed. This work presents evidence leading to the first discovery of a photosystem-related gene involved in regulating fruit flesh coloration.
