Multi-omics analysis and integrative approach to evaluate pharmaco-clinical efficacy of a novel combination of active ingredients on senescence and causes of aging

Skin aging is the consequence of intrinsic and extrinsic factors. At the cellular level, accumulation of senescent cells (SC) is also involved in skin aging. The secretome of SC (SASP), is a cocktail of mediators (chemokine, oxylipin…) involved in skin aging. The reduction of SASP by senotherapeutics is a promising strategy to reduce skin aging. Our aim was to study, by using our interactomic expertise, the senomorphic and anti-aging properties, of a unique cosmetic formulation composed of, among others, 6% Niacinamide, 0.1% High Molecular Weight Hyaluronic Acid (HMW HA) and 0.1% HA Fragment intermediate (HAFi). We developed an original pharmaco-clinical approach based on multi-omics: transcriptomic, metabolomic and metagenomic. We included 30 subjects between 42 and 55 years old (application once a day, for two months). We demonstrated that the cosmetic formulation regulated mRNA, miRNA and metabolites networks, and positively modulate skin microbiota diversity, correlated with skin aging signs. We also measured a senomorphic effect by decreasing SASP-mediatorslike (CXCL9 and Oxylipin). These benefits were associated with improved clinical sign of aging: wrinkles, firmness and plump. Altogether, these results showed for the first time that a cosmetic formulation was able to target cellular senescence and to reduce signs of aging.
