Regulation of effector gene expression as concerted waves in Leptosphaeria maculans: a two-players game

Résumé

Leptosphaeria maculans is a phytopathogenic fungus responsible for the stem canker of oilseed rape and displays a particularly complex lifecycle. As for other fungal parasites, disease establishment-related genes are expressed during infection among which effector-encoding genes. Effectors are molecules involved in the manipulation of host immune response to set up disease. As a set of effector-encoding genes are present within AT- and repeat-rich regions in L. maculans genome, we investigated in the past few years the importance of chromatin remodeling in the virulence of this fungus. We could demonstrate that the repressive histone modification H3K9me3 deposited by the methyltransferase KMT1 is involved in the regulation of genes highly expressed during infection, including effector-encoding ones. Nevertheless, inactivation of KMT1 did not induce expression of these genes at the same level as observed during infection of oilseed rape, suggesting that a second layer of regulation, such as a transcription factor (TF), might be involved. We recently investigated the involvement of Pf2, a TF belonging to the fungal specific Zn2Cys6 family, in the control of effector-encoding genes and showed its concerted role in the regulation of pathogeny with KMT1. Notably, transcriptomic analyses revealed an enhanced effect of the over-expression of LmPf2 on effector gene expression in a ∆kmt1 background, thus unraveling a dual layer control of pathogenesis.
