Validacion del radioinmunoensayo para la estimacion de testosterona, 11-cetotestosterona y 17β-estradiol en plasma de lubina (Dicentrarchus labrax L.)

In the last years, radioimmunoassay (R.I.A.) has been used in order to measure plasma steroid levels. The present work validates R.I.A. for determination of testosterone, 11-ketotestosterone and 17 beta -estradiol in the plasma of sea bass. Prior assay and to increase the assay specificity, the plasma steroids were extracted and purified on Sephadex LH-20 columns. The recoveries (testosterone: 85.2%, 11-ketotestosterone: 88.7%, and 17 beta -estradiol: 71.6%) after chromatography were satisfactories. Precipitation with PEG was used to separate the steroid-antibody complex from free steroid, showing very low levels of non-specific counts. The assay showed a good sensibility, specificity and precision; its accuracy demonstrated that R.I.A. is a valid technic for the measurement of plasma testosterone, 11-ketotestosterone and 17 beta -estradiol in the sea bass
