Indirect involment of tryptophan derivatives, NO- and NO2 in Amyotrophic Lateral Sclerosis, Alzheimer syndrome and Multiple Sclerosis.

Résumé

Ours previous works have described circulating antibodies in Multiple Sclerosis (MS). These antibodies with specific binding, were directed against small endogenous compounds modified or not by radical species. We reported the binding of circulating antibodies in Amyotrophic Lateral Sclerosis (ALS), Alzheimer disease (AD), Remitting Relapsing MS (RR-MS) and Secondary Progressive MS (SP-MS) patient sera. For this purpose, in order to mimic the antigenic modifications induced during the pathogenic processes induced during these neurodegenerative diseases, tryptophan derivatives (TD) conjugates and NO and NO2-modified amino acid conjugates were synthesized.The TD antigens: kynurenin, 3OH-kynurenin, kynurenic acid, picolinic acid, xanthurenic acid, anthranilic acid, 3OH-anthranilic acid, quinolinic acid and quinaldic acid, were linked each of them to bovine serum albumin (BSA) using carbodiimide coupling reaction. NO- and NO2-conjugates resulted of the following compounds: nitrotyrosine, tyrosine, phenylalanine, citrulline, tryptophan, asparagine, creatin, cysteine, methionine, histidine, arginine, linked to BSA via glutaraldehyde reaction and nitrosylated. All conjugates were assayed by adapted ELISA tests. Our studies have shown that: 1) specific circulating antibodies directed against TD, NO- and NO2- modified components were found for each pathology with a specific pattern; 2) specific isotype profiles of immunoglobulins were also observed for each disease.Considering the previous antigens, for ALS, IgG, M, A were principally found. For AD and SP-MS, principally, IgA were found. Conversly for RR-MS, IgM were identified.The identification of IgA and/or IgG or IgM directed against specific antigens underlines the inducible NOSynthase activation and the indolamine2,3-dioxygenase/tryptophan hydroxylase pathway induction
