Role of 50 Hz magnetic fields on ALS development in SOD-1 rats

Background: Epidemiological studies suggest that occupational exposure to electromagnetic fields is associated with ALS (Gunnarsson et al 1991; Davanipour et al 1997; Savitz et al 1998a, 1998b; Johansen &Olsen 1998b, Hakansson et al, 2003). Beside further well-designed epidemiological studies, confirmatory results from additional biological studies specifically designed for the purpose would be valuable. Objectives : The objective of the study is to evaluate, for the first time, if exposure of SOD-1 rats to 50 Hz magnetic fields is able to alter ALS development. Methods: This study is performed on SOD-1 rats (Taconic Europe , Denmark), and is aimed at testing three exposure levels to 50 Hz magnetic fields (10, 100 et 1000 μT). 8 rats per group will be exposed to magnetic fields in Helmholtz coils, beginning before the onset of clinical signs of ALS. Exposure will last 2 hours/day, 5 days/week for 7 weeks. Body weight, survival and motor coordination will be followed. A systematic search will be performed for the presence of new antigenes, that were previously identified in sera from ALS patients and identified mainly as the products of radical processes (for instance NO-cysteine, N-acetyl cysteinyl dopamine, short-chain fatty acids) (Salauze et al, 2005). Sham-exposed rats (placed in non-fed Helmholtz coils) and cage control rats (maintained in the animal facility) will be included in this protocol. Results: Results will be presented at the meeting. The data obtained should provide additional information that will complement the results of epidemiological studies for the risk assessment of neurodegenerative diseases in populations occupationally exposed to electromagnetic fields.
