Mechanical response of a megakaryocyte acoustically coupled with a tethered microbubble

Abstract

In recent years, a growing interest arised for using cavitation microbubbles oscillating in steady state for purposes of sonoporation [1]. The mechanisms commonly proposed for cell membrane permeabilization are the creation of liquid flows (microstreaming) and induced shear stresses on the cell membrane [2], as well as significant cell membrane deformation through push/pull phenomena [3]. We here propose an experimental investigation of the interaction between a wall-attached microbubble and a biological cell. The experimental setup consists of a tank filled with cell culture medium. A megakaryocyte (MK, bone marrow cell) with a mean radius ranging between 20 and 30 μm is maintained by a glass capillary. By means of a micromanipulator, a single MK is mechanically coupled with a single wall-attached oscillating microbubble of comparable size, driven by an ultrasound field. An ultra-fast camera captures the time- resolved description of the bubble-cell interaction at a micro-scale level, and hence the characterization of the cell membrane deformations and the bubble interface oscillations. Then, a broad parametric study aims the pairing up of the sonoporation success (by particles internalization) with specific regime of bubble oscillation and modal deformations.
